Utveckling av en PCR metod för identifiering av nyupptäckta mjölksyrabakterier

Flera olika arter av mjölksyrabakterier som ingår i släktena Lactobacillus och Bifidobacterium har hittats hos bin och i deras honung. Idag finns ingen effektiv metod för identifiering av bakterierna. Syftet med detta projekt är att utveckla en metod för snabb identifiering genom att hitta lämpliga primers till olika mjölksyrabakterier och därmed få fram en Polymeraskedjereaktion (PCR) metod. Ribosomal ribonukleinsyra (rRNA) generna eller 16S-23S rRNA intergenic spacer region (ISR) används ofta vid design av primers, som därefter används i PCR för att identifiera olika bakterier. Deoxiribonukleinsyra (DNA) visualiseras i agarosgelen med hjälp av SYBRgreen I som fluorescens på ultraviolett (UV)-ljusbord.

Optimization and validation of a triplex real-time PCR assay for thermotolerant Campylobacter species associated with foodborne disease

The genus Campylobacter is globally recognised as the leading bacterial cause of human foodborne gastroenteritis. Every year around 8000 Swedes are infected by Campylobacter. Most people are infected by thermotolerant Campylobacter species, commonly C. jejuni and C. coli.

Identifiering av 13 nya mjölksyrabakterier med DHPLC

Mjölksyrabakterier tillhörande släkten Lactobacillus och Bifidobacterium har nyligen upptäckts hos bin och i honungen de producerar och innefattar 13 nya arter[1]. Forskarna arbetar med att ta fram nya snabba och mer pålitliga identifierings metoder för att karakterisera dessa bakterier. I detta projekt undersöktes möjligheten att identifiera dessa bakterier med en ny metod som heter denaturing high performance liquid chromatography (DHPLC). Metoden bygger på separation av PCR (polymerase chain reaction) amplifierade 16S rDNA fragment i DHPLC [8]. Vid identifiering av bakterier amplifierades olika variabla regioner från 16S rRNA genen, som påvisade efter sekvensering störst genetisk variation mellan dessa bakterier [1]. Separationen utfördes med ion-pair reverse-phase high presure liquid chromatoghaphy (IP RP HPLC) med delvis denaturering av DNA molekylen.

Undersökning av potentiella probiotiska organismer i tarmmikroflora hos häst med speciell hänsyn till Lactobacillus spp. :

The aim of this study was to isolate and identify Lactobacillus species indigenous to the gastrointestinal microflora of Swedish horses. These lactobacilli are to be used as probiotic candidates in research concerning antibiotic-associated diarrhoea in horses. Eight horses, age 6-10 years, were selected from a riding school for collection of faeces samples. Man Rogosa Sharp medium and gram staining were used for the selection of supposed lactobacilli. We could identify 33 isolates of supposed lactobacilli. With the use of API tests 22 of these 33 isolates could be identified.

Granulocytär anaplasmos hos hund

Granulocytär anaplasmos orsakas av den gramnegativa bakterien Anaplasma phagocytophilum, som sprids via fästingarten Ixodes ricinus. Det har visats att denna fästingart ökat och spridits norrut i Sverige, där den tidigare inte funnits i lika stor utsträckning på grund av ett för kallt klimat. Orsaken till spridningen är troligen klimatförändringar, då Sveriges medeltemperatur stigit under en längre period. Sjukdomen kan drabba många av våra husdjur men även människan. De vanligaste kliniska symptom som kan uppkomma är feber, dålig aptit och slöhet.

Effects of peat and wood shavings as bedding on the faecal microflora of horses

The main purpose of this study was to determine the impact of bedding material on the faecal microflora of horses. Another objective was to investigate the hygienic and physical qualities of the bedding materials used and how these may change in the course of the trial period. The bedding materials used were sphagnum peat and wood shavings. Six horses had each material as bedding for a three week period in a change?over experiment.

The Bovine respiratory syncytial virus (BRSV) causes bronchiolitis and interstitial pneumonia, predominantly in calves, and is a major cause of bovine respiratory disease worldwide. In humans, BRSV is paralleled by the closely related Human respiratory syncytial virus (HRSV), an important cause of respiratory disease, most severe in infants.The clinical signs and pathology during RSV infection is caused, not only by the direct effects of viral replication, but also by the response of the host immune system. The immunopathology of RSV has long obfuscated our understanding of the disease, and development of effective treatment and vaccines will be very difficult until greater knowledge is gained.One of the components of the immune system that has come into focus in RSV research the last few years, is the Toll-like receptor 4 (TLR4). The TLR4 receptor is well known as the receptor that binds lipopolysaccaride (LPS), and initiates the host response to bacterial infection. Recently, it has been shown that the fusion protein of RSV also interacts with, and up-regulates the expression of, the TLR4 receptor.

Förekomst av Chlamydia/Chlamydophila spp hos gris med och utan konjunktivit.

ABSTRACTChlamydia and Chlamydophila are species of a family of bacteria (Chlamydiaceae) that cause a range of diseases, including conjunctivitis, in a variety of animals including pigs and humans. Some species cause zoonotic disease. This study examined the occurrence of Chlamydiaceae in samples taken from the lower eyelid conjunctiva in pigs with and without conjunctivitis. The study was initiated based on a herd investigation aimed to examine the experienced increase of conjunctivitis in some finisher herds. This study was conducted as a case-control study, by collecting samples from 62 case pigs (16-26 weeks old) and an equal number of controls, from three herds in Uppland, Sweden.

Studie av tarmflorans sammansättning och årstidsvariation i grovtarmen hos travhästar med grovfoderdiet

Syftet med projektet var att undersöka årstids- och den individuella variationen av normala tarmfloran hos travhästar utfodrade med en grovfoderdiet. Hästar är grovtarmsjäsare med ett komplex mikrobiellt ekosystem anpassat för betning av foder med högt fiberinnehåll och lågt energiinnehåll. För att i framtiden kunna veta och jämföra hur olika fodermedel påverkar och förändrar det mikrobiella ekosystemet i hästens mag- och tarmkanal är det av intresse att även veta hur ekosystemet ser ut hos hästar utfodrade med ett naturligt anpassat foder. En större förståelse om hästens biologi och tarmflora är väsentligt att veta för att undvika och behandla tarmsjukdomar. Från 16 stycken hårt tränade travhästar har träck- och foderprov samlats under ett helt år. Vid 9 tillfällen har prover analyserats från alla 16 hästar för att studera mikrobiotans årstidsvariation samt vid 3 tillfällen för 4 av hästarna har prover samlats in under tre efterföljande dagar och analyserats för att studera mikrobiotans dagsvariation.

Lactobacilli in the gastrointestinal tract of dog and wolf : isolation, identification and characterization of L. reuteri

Since the first wolf was taken as a companion animal a lot have happened with the biology of these animals. Is it possible that changes in the gut microbiota of the domestic dog are involved in the increasing numbers of animals with gastrointestinal disorders? Probiotics have been proven to promote the immune system of dogs by increasing concentrations of neutrophils and monocytes, decreasing the fragility of erythrocytes and increasing the serum IgG concentrations. Probiotics have also been shown to improve the fecal consistency, fecal dry matter and defecation frequency of dogs. The aim of this study was to find out if the bacterial counts in the gut of dogs differed from the gut of wolves, with the focus on lactobacilli.

Glycerol to dairy calves : effects on intestinal health and fluid balance

The bacterium Lactobacillus reuteri, which natural habitat is in the intestine of mammals and birds, uses glycerol as a substrate for production of the antimicrobial compound reuterin. Glycerol has been shown to decrease the number of Escherichia coli in human feces and it is believed to be due to in situ production of reuterin. E. coli belongs to the large family of Enterobacteriaceae, naturally occurring in the intestine, and pathogenic strains of E. coli have been shown to be one of several bacteria causing diarrhoea in calves.
